Evaluate the efficacy and safety of tianeptine sodium(CAS 30123-17-2) and paroxetine in the treatment of depression

Objective: To evaluate the efficacy and safety of tianeptine sodium(CAS 30123-17-2) and paroxetine in the treatment of depression. Methods: We searched Chinese Journal Full-text database VIP, Wanfang database, Pub Med, Medline, Em Base regarding tianeptine sodium and paroxetine in the treatment of depression randomized controlled trial (RCT), Cochrane system evaluation method evaluation, Meta-analysis Rev Man5.0 statistical software. Results: A total of eight studies, 936 patients. Meta analysis showed that patients tianeptine sodium significant efficiency, Hamilton Depression Scale (HAMD) score and paroxetine group showed no significant difference (P> 0.05); the incidence of adverse reactions, the two groups was statistically significance (P <0.01). Conclusion: tianeptine sodium(CAS 30123-17-2) and paroxetine in the treatment of depression as effective as, and less adverse reactions.
Effect of chronic stress on tianeptine sodium(CAS 30123-17-2) thiophene serum corticosterone and hippocampal neurons proliferation: Chronic stress leads to serum corticosterone concentration, hippocampal neurons and changes in cell proliferation antidepressant mechanism of tianeptine sodium. Methods: 24 rats were randomly divided into: control group, stress group, stress group with water, stress dosing group (n = 6) were given three weeks unpredictable stress, stress plus drug group were 50mg / kg administered thiophene Kneipp Tingjia 0.9% sodium chloride solution was fed, were given the same amount of water stress in the same proportion 0.9% sodium chloride solution orally, by enzyme-linked immunosorbent assay serum corticosterone concentration, Ki67 immunohistochemistry measuring hippocampal dentate gyrus cell proliferation. 
Results: (1) the difference in the concentration of corticosterone stress before each group was not statistically significant, stress group 1 week corticosterone concentrations low compared with other groups, the difference was statistically significant (P <0.01); the first two weeks of stress corticosterone concentration in the control group compared with other groups lower, the difference was statistically significant (P <0.01), corticosterone stress dosing group was significantly lower than should be water stress and shock group, the difference was statistically significant (P <0.01); the first three weeks of stress and stress-control dosing group corticosterone concentration was significantly lower than the other two groups, the difference was statistically significant (P < 0.01) hippocampus (2) stress dosing rats Ki67 positive cells was significantly higher than water stressed and stress group, the difference was statistically significant (P <0.01). Conclusion: chronic stress can be unpredictable cause corticosterone concentration and decreased hippocampal cell proliferation, tianeptine sodium(CAS 30123-17-2) can prevent or reverse the above changes.
